I use a SD card for speed boost, but wondering if I put a card or USB flash drive in other inputs, will it get faster? In other words if I use 2-3 USB Inputs for SpeedBoost, will it be even fast than just one?

Probably not. SpeedBoost doesn't really speed up your PC. It handles some caching. Adding more is not going to yield any noticeable benefit, in fact, forcing Vista to manage multiple caches may actually slow it down.
